TRITON-PN: A Study to Evaluate the Efficacy and Safety of Nucresiran in Patients With Hereditary Transthyretin Amyloidosis With Polyneuropathy
This research study, called TRITON-PN, is looking into a new medicine named Nucresiran for people who have a condition called hereditary transthyretin amyloidosis (hATTR) that causes nerve damage, known as polyneuropathy. The main goals are to understand how safe Nucresiran is and how well it works. Researchers want to see if it can help improve nerve issues, overall quality of life, how well people can move around, and their general health. They will compare Nucresiran to another existing treatment, Vutrisiran, by checking the levels of a specific protein (transthyretin) in the blood. This is a Phase 3 study, meaning it's one of the final stages of testing before a medicine could potentially be made widely available.

¿De qué trata este estudio?
This study is for people living with a rare inherited condition called hereditary transthyretin amyloidosis, or hATTR for short. In hATTR, a protein called transthyretin (TTR) doesn't fold correctly and builds up in different parts of the body, including the nerves. When it affects the nerves, it's called polyneuropathy, which can lead to problems like numbness, tingling, weakness, and pain.
The main purpose of this study is to find out if a new medicine, Nucresiran, is safe and effective in treating people with hATTR and nerve damage. Researchers will be looking at several things to judge its effectiveness. They will check if it helps improve nerve function, how it impacts your daily life and overall well-being, and if it makes a difference to your ability to move and walk. They will also compare Nucresiran to another medicine, Vutrisiran, that is currently used to treat this condition, specifically by looking at how much of the faulty TTR protein is in the blood.
Taking part in a study like this helps doctors learn more about new treatments. If Nucresiran proves to be safe and helpful, it could become another option for people living with hATTR polyneuropathy in the future. The study is for adults aged 18 to 85, and both men and women can participate.
Puntos clave
- This study is testing a new medicine, Nucresiran, for hATTR polyneuropathy.
- It aims to see if Nucresiran can improve nerve problems and quality of life.
- The study will compare Nucresiran with an existing treatment, Vutrisiran.
- Participation involves regular clinic visits and health checks.
- You can discuss if this study is right for you with your doctor.
¿Quién puede ser elegible?
To join this study, you would need to have been officially diagnosed with hATTR that causes nerve damage (polyneuropathy) and have a specific change in your TTR gene. Your nerve problems should be within a certain range of severity, and you should generally be able to manage your daily activities.
There are also some reasons why you might not be able to join. For example, if you've had a liver transplant or are likely to need one soon, you wouldn't be eligible. The study also can't include people who have other types of amyloidosis, severe heart failure, or certain problems with their liver or kidneys. If you have other known causes for nerve problems that aren't related to hATTR, you also wouldn't be able to take part.
These rules are in place to help make sure the study is safe for everyone involved and that the results are clear and accurate. Your study doctor would carefully check all these points with you.

Responde estas preguntas rápidas para ver si puedes ser elegible. Esto es solo una guía: el equipo de investigación toma la decisión final.
- I have been diagnosed with hATTR that causes nerve problems.
- My nerve problems are not too mild or too severe.
- I am generally able to carry out my daily activities.
- I haven't had a liver transplant.
- I don't have other serious heart, liver, or kidney conditions.
¿En qué consiste la participación?
If you decide to take part in this study, you will be given one of the study medicines, either Nucresiran or Vutrisiran. You will need to attend regular visits to the study clinic over a period of time. These visits will involve various assessments to check your health and how the medicine is affecting you. This might include physical exams, blood tests, urine tests, checks of your nerve function, and questions about your quality of life and how you're feeling. You'll be closely monitored by the study team throughout. The total duration of your participation in the study, including follow-up, would be fully explained to you before you agree to join.
